Overview

QTX3544 + cetuximab is a **combination therapy** currently being evaluated in clinical trials for KRAS G12V-mutated solid tumors. QTX3544 is an oral, allosteric, G12V-preferring, multi-KRAS inhibitor developed to target the oncogenic KRAS G12V mutation, which drives tumorigenesis via sustained activation of the MAPK signaling pathway. Preclinical data demonstrate that combining QTX3544 with **cetuximab** (an EGFR inhibitor monoclonal antibody) results in synergistic anti-tumor effects: QTX3544 locks KRAS mutants in an inactive conformation, preventing effector signaling and protein-protein interactions, while cetuximab blocks EGFR and further suppresses downstream MAPK signaling. This combination leads to enhanced and more durable tumor regression in models of pancreatic and colorectal cancer cell lines driven by KRAS G12V[1][2][3][4][5][6][7].
