Overview

Quabodepistat (OPC-167832) is a novel small molecule carbostyril derivative developed as an anti-tuberculosis agent. It acts by inhibiting decaprenylphosphoryl-β-D-ribose 2'-oxidase (DprE1), an essential enzyme for the biosynthesis of the cell wall in Mycobacterium tuberculosis. This mechanism disrupts the formation of arabinans in the bacterial cell wall, leading to potent bactericidal activity against both growing and intracellular bacilli. Quabodepistat has demonstrated strong efficacy in preclinical models and early clinical trials against drug-susceptible and multidrug-resistant/extensively drug-resistant tuberculosis strains at low concentrations. It is being investigated primarily for use in combination regimens with other new anti-TB drugs such as delamanid and bedaquiline to potentially shorten treatment duration for pulmonary tuberculosis[1][2][4][7].
