**QUAD 3.0-WP936** is an investigational, site-specifically conjugated multivalent protein-drug conjugate for glioblastoma. Its QUAD 3.0 targeting protein combines an IgG1 scaffold, ephrin-A5, and the IL-13.E13K mutant to bind ephrin type-A receptor 2, ephrin type-A receptor 3, ephrin type-B receptor 2, and interleukin-13 receptor subunit alpha-2 across the glioblastoma tumor microenvironment. The construct carries WP936, a thiol-reactive modified doxorubicin payload, to deliver a DNA-binding cytotoxic agent after tumor-cell binding and internalization. It showed low-nanomolar cytotoxicity in established and patient-derived glioblastoma cell lines and was evaluated by local convection-enhanced delivery in animal glioma models.
