Overview

The quadrivalent recombinant dengue vaccine (DSV4) is a virus-like particle (VLP)-based vaccine candidate designed to provide broad protection against all four serotypes of the dengue virus (DENV-1, DENV-2, DENV-3, and DENV-4). Developed by the International Centre for Genetic Engineering and Biotechnology (ICGEB) in collaboration with Sun Pharmaceutical Industries, the vaccine utilizes a recombinant fusion protein architecture. It incorporates the envelope domain III (EDIII) from each of the four dengue serotypes, which are fused to the Hepatitis B virus surface antigen (HBsAg). The HBsAg component acts as a scaffold that drives the self-assembly of the fusion proteins into VLPs, presenting the dengue antigens in a highly organized, repetitive array to the immune system. This approach is intended to induce potent neutralizing antibodies while minimizing the risk of antibody-dependent enhancement (ADE) by excluding the highly cross-reactive prM and fusion loop epitopes found in whole-virus vaccines.
