Overview

Quetiapine + venlafaxine is a combination of two pharmaceutical drugs used primarily in the management of treatment-resistant depression (TRD) and other mood disorders. Quetiapine is an atypical antipsychotic that acts as an antagonist at serotonin 5-HT2, dopamine D1 and D3, and histamine H1 receptors. Venlafaxine is a serotonin-norepinephrine reuptake inhibitor (SNRI) that increases synaptic concentrations of serotonin and norepinephrine by inhibiting their reuptake. The combination has been studied for its potential to enhance antidepressant efficacy in patients who do not respond adequately to monotherapy, with evidence suggesting improved outcomes compared to either agent alone[1][4]. Both drugs are small molecules administered orally. The combination may increase the risk of certain side effects such as metabolic syndrome or cardiovascular effects, so monitoring is recommended[3]. Pharmacokinetic interactions have been observed due to shared hepatic metabolic pathways, particularly involving CYP3A4[2].
