Conducted by Cliniques universitaires Saint-Luc, the Phase 1 study investigated the impact of these agents on insulin sensitivity, pancreatic beta-cell function, and adiponectin levels in lean, drug-naïve patients with schizophrenia over a 9-month period. While all four drugs are FDA-approved for the treatment of schizophrenia and bipolar disorder, they possess distinct pharmacological properties and varying propensities for causing weight gain and metabolic syndrome. Olanzapine is typically associated with the highest risk of metabolic disruption, whereas aripiprazole is considered relatively metabolic-neutral due to its unique partial agonist activity at dopamine receptors.
