QUILT-3.076 is an investigational combination immunotherapy regimen developed by ImmunityBio for the treatment of locally advanced or metastatic solid tumors. The regimen consists of autologous memory cytokine-enhanced natural killer (M-ceNK) cells co-administered with nogapendekin alfa inbakicept (N-803, ANKTIVA), a first-in-class interleukin-15 (IL-15) superagonist. M-ceNK cells are patient-derived natural killer cells that are pre-activated ex vivo with a cytokine cocktail (including IL-12, IL-15, and IL-18) to induce a highly cytotoxic, memory-like phenotype characterized by elevated interferon-gamma (IFN-gamma) and granzyme B production. Nogapendekin alfa inbakicept, which consists of an IL-15 mutant fused to an IL-15 receptor alpha-Fc complex, acts as an agonist of the IL-2/IL-15 receptor beta-gamma heterodimer on NK and CD8+ T cells. This superagonist is designed to promote the in vivo proliferation, persistence, and anti-tumor activity of the adoptively transferred M-ceNK cells. The combination is currently being evaluated in a Phase 1 clinical trial (NCT04898543) for patients with advanced or metastatic solid tumors.
