quisinostat + proteasome inhibitors is a small molecule combination therapy being developed by NewVac for the treatment of translocation-associated sarcomas. The combination pairs quisinostat, a potent and orally bioavailable histone deacetylase 1 (HDAC1) inhibitor, with proteasome inhibitors to induce apoptosis in tumor cells. This therapeutic strategy specifically targets fusion oncoprotein complexes characteristic of these rare sarcomas, such as SS18/SSX in synovial sarcoma, EWS/ATF1 in clear cell sarcoma, FUS/DDIT3 in myxoid liposarcoma, and EWS/WT1 in desmoplastic small round cell tumor (DSRCT). By inhibiting both HDAC1 and the proteasome, the treatment aims to disrupt the stability and function of these oncogenic drivers, which are otherwise difficult to target directly.
