Quizartinib blocks signaling activity of FLT3, a receptor tyrosine kinase frequently mutated in AML, thereby inhibiting leukemic cell proliferation and inducing apoptosis. Cytarabine acts as an antimetabolite that incorporates into DNA, disrupting DNA synthesis and cell division. This combination is used alongside an anthracycline during induction, followed by high-dose cytarabine consolidation and quizartinib maintenance. The regimen is FDA-approved based on phase 3 trials demonstrating increased overall survival in FLT3-ITD positive AML populations[2][3][10].
