Overview

Quizartinib + cytarabine + mitoxantrone is a combination therapeutic regimen being evaluated for the treatment of relapsed or refractory (R/R) acute myeloid leukemia (AML) harboring FLT3-ITD mutations. The regimen, often referred to as Q-HAM in the context of the clinical trial NCT03989713 led by Prof. Dr. Richard F. Schlenk, combines the targeted FLT3 inhibitor quizartinib with high-dose cytarabine and mitoxantrone salvage chemotherapy. Quizartinib is an oral, highly selective, second-generation small molecule inhibitor of the FLT3 receptor tyrosine kinase, developed by Daiichi Sankyo and marketed as Vanflyta. Cytarabine is an antimetabolite that interferes with DNA synthesis, and mitoxantrone is an anthracenedione that inhibits topoisomerase II and intercalates into DNA. The Q-HAM trial specifically explores the efficacy of this combination followed by either prophylactic or minimal residual disease (MRD)-triggered maintenance therapy with quizartinib.
