QXL138AM is a first-in-class masked immunocytokine developed by Nammi Therapeutics for the treatment of multiple myeloma and various solid tumors. The molecule is a recombinant fusion protein that combines a monoclonal antibody targeting CD138 (Syndecan-1) with an attenuated or "masked" version of interferon-alpha 2 (IFNα2). CD138 is highly expressed on the surface of multiple myeloma cells and several types of solid tumor cells, making it an effective anchor for targeted delivery. By masking the interferon moiety, QXL138AM remains relatively inactive in the systemic circulation, which is intended to reduce the severe side effects typically associated with systemic interferon therapy. Upon binding to CD138 on the tumor cell surface, the interferon is positioned to activate interferon receptors (IFNAR1/2) on the tumor cells and adjacent immune cells, promoting anti-tumor activity through direct growth inhibition and immune system stimulation. QXL138AM is currently undergoing Phase 1a/1b clinical evaluation to assess its safety, pharmacokinetics, and preliminary efficacy in patients with advanced malignancies.
