QYK-101 is an experimental **in situ vaccine** and **Toll-like receptor 7/8 (TLR7/8) agonist** being developed and investigated by **Nanjing Drum Tower Hospital**. It is designed for **intratumoral injection** to stimulate the local innate immune system within the tumor microenvironment. By activating TLR7 and TLR8 on antigen-presenting cells, such as dendritic cells, QYK-101 promotes their maturation and the subsequent presentation of tumor-associated antigens to T cells. This mechanism is intended to convert immunologically "cold" tumors into "hot" tumors, fostering a systemic antitumor immune response. In clinical trials, QYK-101 is frequently evaluated in combination with **hypofractionated radiotherapy**, which facilitates the release of tumor antigens through immunogenic cell death, thereby potentially inducing an abscopal effect against distant metastatic lesions.
