Overview

R-7050 is a cell-permeable small molecule that acts as a selective inhibitor of Tumor Necrosis Factor Receptor 1 (TNFR1) signaling. It functions by preventing the internalization of the TNFR1 complex, a critical step for the activation of downstream pro-inflammatory signaling cascades, including the NF-κB and MAPK pathways. Unlike traditional TNF-α inhibitors that neutralize the cytokine itself, R-7050 specifically targets the receptor's intracellular trafficking and signaling assembly. Preclinical studies have highlighted its potential in mitigating neuroinflammation, reducing edema, and preserving blood-brain barrier integrity in models of intracerebral hemorrhage and ischemic stroke. It is primarily utilized as a high-affinity pharmacological tool in biochemical research to dissect TNFR1-mediated biological processes.
