R-DOTAP (Versamune) is an enantiospecific cationic lipid nanoparticle that serves as a potent immune-activating adjuvant and immunotherapy agent. Unlike the racemic mixture DOTAP, the R-enantiomer specifically triggers the activation of dendritic cells and induces type I interferon responses. Its mechanism of action involves signaling through endosomal Toll-like receptor 7 (TLR7) and Toll-like receptor 9 (TLR9) in a MyD88-dependent manner, leading to the induction of robust CD8+ and CD4+ T-cell responses. Developed by PDS Biotechnology, R-DOTAP is utilized as the core component of the Versamune platform to deliver antigens and stimulate the immune system against various cancers, including HPV-associated malignancies, and infectious diseases like COVID-19 and influenza.
