**r-methadone + TAAP** is a novel combination prodrug consisting of the R-enantiomer of methadone chemically linked to a Trypsin Activated Abuse Protection (TAAP) moiety. The TAAP technology is designed to make r-methadone active only after enzymatic activation in the small intestine (by trypsin), significantly reducing misuse and overdose potential by preventing effective drug release via alternate routes of administration (e.g., crushing, snorting, injecting). The prodrug may also be combined with MPAR (multi-pill abuse resistance) to control oral multi-dose pharmacokinetics. The underlying goal is to provide effective opioid use disorder (OUD) treatment with minimized abuse and overdose risk. Developed as a next-generation methadone for OUD, it is intended to advance through clinical development for prevention and treatment of opioid dependence and overdose[1].
