R-Star001 is an autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Celconta (Guangdong Xiankangda Biotechnology) for the treatment of Nectin-4-positive advanced solid tumors. The CAR-T cells are engineered to target **Nectin cell adhesion molecule 4 (Nectin-4)**, a protein frequently overexpressed in various epithelial malignancies, including urothelial carcinoma, breast cancer, and lung cancer. In addition to the CAR construct, R-Star001 is modified to induce the secretion of **Interleukin-18 (IL-18)**, a pro-inflammatory cytokine. This "armored" CAR-T approach is intended to overcome the immunosuppressive tumor microenvironment, enhance CAR-T cell expansion and persistence, and recruit endogenous immune cells to the tumor site. The drug is currently being evaluated in a Phase 1 exploratory clinical trial (NCT07101549) across multiple solid tumor indications.
