R0.6C is a transmission-blocking malaria vaccine candidate developed by Radboud University Medical Center. It is a recombinant fusion protein consisting of the Pfs48/45 domain 3 (designated 6C) and the N-terminal region of the Plasmodium falciparum glutamate-rich protein (GLURP, designated R0). The vaccine is produced using a *Lactococcus lactis* expression system. Its mechanism of action involves inducing antibodies in the human host that, when ingested by a mosquito during a blood meal, target the Pfs48/45 protein on the surface of the parasite's sexual stages. This interaction blocks the development of the parasite within the mosquito gut, thereby preventing the transmission of malaria to other humans. The vaccine has been evaluated in Phase I clinical trials (STOP-TRANS study) using either Alhydrogel (aluminum hydroxide) or Matrix-M1 as adjuvants.
