R1065 is a preclinical-stage therapeutic candidate developed by Harbour BioMed (HBMK) for the treatment of IgA Nephropathy (IgAN). It is a monoclonal antibody designed to target and neutralize A Proliferation-Inducing Ligand (APRIL), a member of the tumor necrosis factor (TNF) superfamily. APRIL is a key regulator of B-cell maturation and is essential for the production of immunoglobulin A (IgA). In patients with IgAN, the overproduction of galactose-deficient IgA1 (Gd-IgA1) leads to the formation of immune complexes that deposit in the renal mesangium, causing inflammation and kidney damage. By inhibiting APRIL, R1065 aims to reduce the levels of pathogenic IgA1, thereby addressing the underlying cause of the disease and preserving renal function.
