Overview

R107 is an investigational, oral, extended-release formulation of racemic ketamine. Developed by Douglas Pharmaceuticals in collaboration with Zenith Technology, it is intended for the treatment of treatment-resistant depression (TRD) and various anxiety disorders, such as social anxiety disorder (SAD) and generalized anxiety disorder (GAD). As an NMDA receptor antagonist, ketamine works by modulating glutamatergic neurotransmission. The R107 formulation is specifically designed to release the active ingredient slowly, aiming to reduce the peak plasma concentrations that cause dissociative and cardiovascular side effects, thereby allowing for potential at-home administration. In the Phase 2 BEDROC trial, R107 demonstrated a statistically significant reduction in depression scores compared to placebo.
