Overview

**R1507 + bevacizumab + carboplatin + paclitaxel** is an investigational combination regimen consisting of four agents: - **R1507**: a fully human monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), designed to inhibit IGF-1R-mediated signaling pathways important for tumor cell growth and survival. - **Bevacizumab**: a recombinant humanized monoclonal antibody targeting and neutralizing vascular endothelial growth factor A (VEGF-A), thereby inhibiting angiogenesis crucial for tumor vascularization and growth. - **Carboplatin**: a platinum-based cytotoxic chemotherapy that induces DNA cross-linking, leading to apoptosis in rapidly dividing cells. - **Paclitaxel**: a cytotoxic agent that stabilizes microtubules, preventing cell division and leading to apoptosis. The combination is developed and studied primarily for advanced solid tumors, particularly ovarian, lung, and other cancers where anti-VEGF and cytotoxic chemotherapy have shown synergistic efficacy. R1507’s IGF-1R targeting is experimental and not yet approved for clinical use; bevacizumab (Avastin), carboplatin, and paclitaxel are FDA-approved. This quadruple regimen is investigational and has appeared in phase I–II trials, with the backbone of bevacizumab, carboplatin, and paclitaxel supported by extensive randomized data for ovarian and lung cancers[2][3][4][5][6][7][8].
