Overview

This is a **combination therapy** consisting of R1507 (a fully human monoclonal antibody targeting Insulin-like growth factor-1 receptor [IGF-1R][1][2][5][8]), **capecitabine** (a prodrug of 5-fluorouracil, acting as an antimetabolite disrupting DNA synthesis), and **trastuzumab** (a monoclonal antibody targeting the Human Epidermal growth factor Receptor 2 [HER2][3][6]). - **R1507** blocks IGF-1R, which plays a role in cell proliferation and survival in various tumors. - **Capecitabine** is orally administered and is converted to 5-FU preferentially in tumors, inhibiting DNA synthesis and cell division. - **Trastuzumab** binds to HER2, inhibiting downstream signaling and flagging cancer cells for immune-mediated destruction. This combination is experimental, studied primarily for certain solid tumors, particularly breast cancer **with HER2 overexpression** and potentially IGF pathway activation, or as salvage therapy in resistant or heavily pretreated settings.
