R1507 + pemetrexed is an **experimental combination therapy** comprising R1507, a monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), and pemetrexed, an FDA-approved antifolate chemotherapy. R1507 inhibits IGF-1R signaling, a pathway implicated in cell proliferation and survival in certain cancers, while pemetrexed inhibits folate-dependent enzymes critical for nucleic acid synthesis, leading to cytotoxicity in rapidly dividing cells. R1507 was developed for oncologic indications but has not advanced to regulatory approval. Pemetrexed is approved primarily for non-squamous non-small cell lung cancer (NSCLC) and malignant pleural mesothelioma, often in combination with other agents. This combination is being or has been investigated in the context of cancer therapy, but it is not approved and remains experimental.
