**R1507 + temozolomide** is an experimental combination therapy comprising R1507, a fully human monoclonal antibody targeting the insulin-like growth factor 1 receptor (IGF-1R), and temozolomide, an oral alkylating agent of the imidazotetrazine class. R1507 acts by selectively inhibiting IGF-1R-mediated signaling, which is implicated in tumor growth and survival. Temozolomide exerts its antitumor effect through DNA methylation, leading to DNA damage and apoptosis in rapidly dividing tumor cells. This combination has been explored in clinical trials targeting solid tumors, particularly central nervous system malignancies and sarcomas, with the rationale that dual targeting of growth signaling and direct DNA damage may provide synergistic antitumor activity.
