Overview

R18 is a high-affinity peptide inhibitor of the 14-3-3 family of proteins. Originally identified through phage display, R18 binds to the conserved amphipathic groove of 14-3-3 proteins, thereby competitively blocking their interaction with various phosphorylated signaling partners. By disrupting these protein-protein interactions, R18 modulates multiple cellular processes, including metabolic reprogramming, cell proliferation, and migration. In preclinical models of pulmonary hypertension (PH), R18 has been shown to reduce pulmonary vascular remodeling, lower right ventricular systolic pressure, and improve hemodynamics by restoring endothelial cell-pericyte communication and inhibiting the pathological proliferative phenotype of pericytes. It is widely utilized as a research tool to study the regulatory roles of 14-3-3 proteins in health and disease.
