Drug intelligence / Profile preview
R21 + AS01B is an experimental malaria vaccine candidate consisting of the R21 virus-like particle (VLP) antigen and the AS01B adjuvant system. R21 is a recombinant protein particle that displays the circumsporozoite protein (CSP) of *Plasmodium falciparum*, designed to elicit high-titer antibodies and T-cell responses against the pre-erythrocytic stage of the malaria parasite. AS01B is a liposome-based adjuvant system developed by GSK, containing two immunostimulants: MPL (3-O-desacyl-4'-monophosphoryl lipid A) and QS-21. This specific formulation was evaluated in a Phase I clinical trial (NCT02600975) by the University of Oxford's Jenner Institute to assess safety and immunogenicity in healthy adults. While R21 was initially tested with AS01B, the program ultimately transitioned to using the Matrix-M adjuvant for later-stage development and eventual regulatory approval.
