R24

R24 is an IgG3 mouse monoclonal antibody designed to target GD3 ganglioside, a prominent antigen found on the surface of melanoma cells and other cells of neuroectodermal origin. In a Phase I clinical trial for patients with metastatic melanoma, R24 demonstrated dose-related peak antibody levels in serum. The drug's mechanism of action involves binding to GD3, which leads to inflammatory reactions, including urticaria, pruritus, erythema, and subcutaneous ecchymoses around tumor sites. Biopsies revealed lymphocyte and mast cell infiltration, mast cell degranulation, and complement deposition, indicating an immune-mediated anti-tumor response. The side effects observed were mild and manageable with antihistamines, and major tumor regression was noted in some patients.
