Overview

R835 is an orally available, potent and selective small molecule inhibitor of interleukin receptor-associated kinase 1 (IRAK1) and IRAK4. Developed by Rigel Pharmaceuticals, it blocks inflammatory cytokine production in response to toll-like receptor (TLR) and interleukin-1 family receptor (IL-1R) signaling. These pathways are critical in the innate immune response, and their dysregulation can lead to a variety of inflammatory conditions. Dual inhibition of IRAK1 and IRAK4 by R835 allows for more complete suppression of pro-inflammatory cytokine release. Preclinical studies have shown activity in rodent models of diseases such as psoriasis, arthritis, lupus, multiple sclerosis, and gout. The drug has been investigated primarily for autoimmune disorders and inflammation[1][2][3].
