R835

R835 is an orally available, potent and selective small molecule inhibitor of interleukin receptor-associated kinase 1 (IRAK1) and IRAK4. Developed by Rigel Pharmaceuticals, it blocks inflammatory cytokine production in response to toll-like receptor (TLR) and interleukin-1 family receptor (IL-1R) signaling. These pathways are critical in the innate immune response, and their dysregulation can lead to a variety of inflammatory conditions. Dual inhibition of IRAK1 and IRAK4 by R835 allows for more complete suppression of pro-inflammatory cytokine release. Preclinical studies have shown activity in rodent models of diseases such as psoriasis, arthritis, lupus, multiple sclerosis, and gout.
