Overview

RA04075215A is a selective and potent small molecule inhibitor of serum- and glucocorticoid-regulated kinase 1 (SGK1) being developed by Sanofi. It is primarily investigated for the treatment of sickle cell disease (SCD) and other beta-hemoglobinopathies. The compound's mechanism of action involves the inhibition of SGK1, which prevents the phosphorylation and subsequent inhibition of the transcription factor FOXO3a. By maintaining FOXO3a in its active state, RA04075215A promotes the expression of fetal hemoglobin (HbF) in erythroid cells. Preclinical studies have demonstrated that treatment with RA04075215A increases HbF protein levels and significantly reduces the sickling of red blood cells under hypoxic conditions, suggesting its potential as a therapeutic agent to alleviate the clinical manifestations of SCD.
