RA16-TD is a targeted DNA-based nanomaterial developed for the treatment of non-small cell lung cancer (NSCLC). It is composed of the RNA aptamer RA16, which exhibits high affinity for NCI-H460 lung cancer cells, conjugated to a tetrahedral DNA (TD) nanostructure. This functionalized nanoparticle addresses the inherent instability of free RNA aptamers in vivo while leveraging the high drug-loading capacity and biocompatibility of DNA tetrahedra. Preclinical studies indicate that RA16-TD significantly enhances cellular uptake and tumor accumulation compared to the free aptamer. When loaded with chemotherapeutic agents such as epirubicin, the construct demonstrates superior therapeutic efficacy and tumor inhibition in xenograft mouse models, suggesting its potential as a versatile platform for targeted NSCLC therapy.
