rAAV-DCN is an investigational gene therapy consisting of a recombinant adeno-associated virus (rAAV) vector designed to deliver the decorin (DCN) gene. Decorin is a small leucine-rich proteoglycan that functions as a natural antagonist of transforming growth factor-beta (TGF-β), a key mediator of tissue fibrosis and inflammation. By overexpressing decorin, rAAV-DCN aims to mitigate pathological remodeling in conditions such as diabetic cardiomyopathy. Preclinical studies indicate that rAAV-DCN treatment can improve left ventricular function and reduce cardiac inflammation and fibrosis by modulating the TGFβ1/Smad and NF-κB signaling pathways. The therapy has also been explored for its potential to regulate other factors such as IGF-IR, PKC-α, and Hsp70 to provide cardioprotective effects.
