**rAAV E7CTL-hsp** is a recombinant adeno-associated virus (rAAV) vector-based therapeutic cancer vaccine encoding a fusion gene construct of the cytotoxic T lymphocyte (CTL) epitope from human papillomavirus type 16 (HPV-16) E7 protein and a heat shock protein (hsp, specifically Mycobacterium tuberculosis hsp70). Delivered as a gene therapy, rAAV E7CTL-hsp is designed to induce a potent antitumor immune response, primarily via the activation of both CD8+ and CD4+ T cells, targeting E7-expressing tumor cells. Mechanistically, the construct targets antigen-presenting cells for both MHC class I and II presentation, resulting in robust CTL and helper T-cell responses leading to elimination of HPV-16 E7-expressing tumor cells. The vaccine has demonstrated efficacy in preclinical murine models for generating tumor elimination and is a potential therapeutic approach for HPV-induced cancers, notably cervical cancer[1][3].
