rAAV-sPD1-TWIST1 is a recombinant adeno-associated virus (rAAV)-vectored gene therapy candidate encoding a fusion protein composed of soluble programmed cell death protein 1 (sPD1) and the TWIST1 antigen. This unique construct delivers simultaneous immune checkpoint blockade via persistent sPD1 production and enhanced antigen-specific T cell responses through vaccination against the tumor-associated antigen TWIST1. The therapy is designed primarily for the treatment of malignant mesothelioma and potentially other TWIST1-expressing tumors, acting through a combination of immune activation, checkpoint inhibition, and local oncosuppression. In preclinical models, rAAV-sPD1-TWIST1 administered intratumorally led to the eradication of established mesothelioma by activating TWIST1-specific cytotoxic T lymphocytes and enhancing immune infiltration into tumors[1][2][5].
