rAAV1-PG9DP is a recombinant adeno-associated virus serotype 1 (rAAV1) vector-based gene therapy designed to deliver the gene encoding PG9, a broadly neutralizing human monoclonal IgG1 antibody targeting the V1V2 loop of the HIV-1 envelope gp120 protein. The therapy aims to induce sustained in vivo expression of PG9 antibody following intramuscular injection, providing immunoprophylaxis against HIV infection. Developed as an alternative to traditional vaccination, this approach leverages AAV-mediated delivery for long-term antibody production. The drug was initially developed by The Children's Hospital of Philadelphia and has been evaluated in phase 1 clinical trials for safety, tolerability, and immunogenicity in healthy adult males[2][3][6].
