Overview

rAAV2-TNFR:Fc is a gene therapy agent consisting of a recombinant adeno-associated virus serotype 2 (rAAV2) vector encoding the cDNA for the human tumor necrosis factor receptor-immunoglobulin Fc fusion gene (TNFR:Fc). This product is designed for intra-articular injection, leading to local expression of the soluble TNF receptor fusion protein in the joint. TNFR:Fc acts as a decoy receptor, binding and neutralizing tumor necrosis factor-alpha (TNF-α), a key pro-inflammatory cytokine involved in the pathogenesis of inflammatory arthritis such as rheumatoid arthritis. Preclinical and early phase clinical studies have demonstrated the safety and tolerability of intra-articular rAAV2-TNFR:Fc, with adverse reactions limited mostly to transient injection site reactions. The primary indication investigated has been inflammatory arthritis, especially rheumatoid arthritis, with studies ongoing or completed in this area[1][2][4][6][7].
