RAB7 siRNA is a small interfering RNA designed to silence the expression of the RAB7 gene, which encodes an endosomal GTPase involved in endocytic trafficking and lysosomal biogenesis. In the context of pulmonary arterial hypertension (PAH) research, RAB7 siRNA is utilized as a laboratory tool to model the deficiency of RAB7 observed in the pulmonary artery endothelial cells (PAECs) of patients. Silencing RAB7 in PAECs leads to endothelial dysfunction, senescence, and an altered secretome that promotes the aberrant proliferation and phenotypic shifting of smooth muscle cells and adventitia fibroblasts. This research tool has been instrumental in identifying downstream effectors, such as the RNA-binding protein CSDE1, which regulates the endothelial secretome under conditions of RAB7 deficiency.
