Overview

Rabepirin (DKP103) is an oral fixed-dose combination (FDC) therapy developed by Dongkwang, consisting of the antiplatelet agent aspirin and the proton pump inhibitor (PPI) rabeprazole. Classified as an 'improved new drug' (incrementally modified drug) in South Korea, it is indicated for the prevention of cardiovascular events in patients who require long-term aspirin therapy but are at high risk for developing gastric or duodenal ulcers. Aspirin provides antithrombotic effects by irreversibly inhibiting cyclooxygenase-1 (COX-1), which prevents the synthesis of thromboxane A2 and subsequent platelet aggregation. Rabeprazole mitigates the gastrointestinal side effects of aspirin by irreversibly inhibiting the H+/K+-ATPase enzyme system (the proton pump) in gastric parietal cells, thereby reducing gastric acid secretion. Rabepirin was approved and launched in South Korea in 2024.
