Overview

ZESPEA-IT is an oral fixed-dose combination capsule developed and marketed by Thrift Pharmaceuticals. It contains enteric-coated rabeprazole sodium (20 mg), a proton pump inhibitor (PPI), and sustained-release itopride hydrochloride (150 mg), a prokinetic agent. Rabeprazole suppresses gastric acid secretion by covalently binding to and inhibiting the gastric H+/K+-ATPase (proton pump) in parietal cells. Itopride enhances gastrointestinal motility and provides antiemetic effects through its dual mechanism of action as a dopamine D2 receptor antagonist and an acetylcholinesterase inhibitor. This combination is primarily indicated for the management of gastroesophageal reflux disease (GERD), functional dyspepsia, and associated symptoms such as heartburn, acid regurgitation, bloating, and nausea.
