Rabeximod is an orally administered small molecule immunomodulator developed primarily for the treatment of moderate to severe active rheumatoid arthritis. It is a synthetic indolo[2,3-b]quinoxaline derivative originally developed by OxyPharma and later acquired by Cyxone. Rabeximod acts as a cell differentiation modulator and macrophage inhibitor, impairing the differentiation of monocytes into pro-inflammatory antigen-presenting cells such as dendritic cells and allostimulated macrophages, while sparing anti-inflammatory macrophages. This leads to reduced immunostimulatory capacity of these cells and decreased severity of arthritis in preclinical models. The drug has completed Phase 1 and Phase 2 trials in rheumatoid arthritis, as well as a Phase 2 trial for COVID-19 infections. Its mechanism involves selective inhibition of pro-inflammatory immune cell function at a fundamental level[1][3][4][5][6][8].
