Rabies virus glycoprotein (RVG)-decorated extracellular vesicles are an engineered nanodelivery platform designed to facilitate the transport of therapeutic agents across the blood-brain barrier (BBB). The system utilizes a 29-amino acid peptide derived from the rabies virus glycoprotein, which exhibits high affinity for the nicotinic acetylcholine receptor (nAchR) expressed on brain capillary endothelial cells and neurons. By functionalizing the surface of extracellular vesicles (EVs) with this peptide, the vesicles can undergo receptor-mediated transcytosis to enter the central nervous system. This technology is primarily investigated as a modular delivery vehicle for various cargos, including small molecules like echinatin, proteins, or nucleic acids, to treat neurodegenerative conditions such as Parkinson's disease by providing targeted neuroprotection and reducing systemic side effects.
