Overview

Racotumomab is an anti-idiotypic mouse monoclonal antibody that functions as a therapeutic cancer vaccine. It mimics the NGcGM3 ganglioside, a tumor-associated neoantigen highly expressed in several human cancers such as non-small cell lung cancer (NSCLC), breast cancer, melanoma, colon cancer, and neuroectodermal pediatric tumors. By acting as an immunogen rather than a passive antibody therapy, racotumomab stimulates the patient's immune system to recognize and attack tumor cells expressing NGcGM3. This mechanism leads to direct cytotoxicity against malignant cells while minimizing cross-reactivity with healthy tissues due to the restricted expression of NGcGM3 in normal human cells. Racotumomab has demonstrated clinical benefit in prolonging survival and reducing metastasis in advanced NSCLC patients and has been studied for other malignancies including glioma, Wilms' tumor, neuroblastoma, retinoblastoma, Ewing's sarcoma, breast cancer, and melanoma[1][2][5][7].
