Overview

rAd-anti-PD-1 is an experimental recombinant adenoviral gene therapy designed to express an anti-programmed death-1 (anti-PD-1) sequence. By utilizing a recombinant adenovirus vector, the drug delivers genetic material to host cells to produce anti-PD-1 locally, which blocks the PD-1/PD-L1 checkpoint pathway. This blockade is intended to rejuvenate exhausted T cells and enhance the immune system's ability to recognize and destroy tumor cells. Preclinical studies in melanoma models have demonstrated that rAd-anti-PD-1 can inhibit tumor growth, promote apoptosis, and induce the maturation of dendritic cells (DCs). Furthermore, it has been shown to increase the production of interferon-alpha and interferon-beta and stimulate the generation of PD-1-specific cytotoxic T lymphocytes (CTLs).
