Overview

rAd-mGM-CSF is a recombinant adenovirus vector engineered to express **mouse granulocyte-macrophage colony-stimulating factor (mGM-CSF)**. It is designed for use in translational cancer immunotherapy and preclinical studies. The vector delivers mGM-CSF locally, where it functions as **a cytokine to stimulate myeloid cell (granulocyte and macrophage) proliferation, differentiation, and activation**, promoting systemic and tumor-local immune responses. The mechanism involves immune modulation, including upregulation of chemokines, infiltration/activation of cytotoxic T cells, and augmentation of anti-tumor immunity. In animal models, rAd-mGM-CSF or related vectors (such as "OncoVEXmGM-CSF" and similar constructs) have been demonstrated to generate strong local and systemic antitumor effects, especially when combined with immune checkpoint inhibitors like CTLA-4 blockade. The drug is investigational, primarily used in research settings for cancer, with reported preclinical efficacy in mouse tumor models[5].
