Overview

**rAd-p53 + chemotherapy** is a combination therapy comprising a gene therapy agent (rAd-p53, also known as recombinant adenovirus-p53 or Gendicine) and one or more standard chemotherapy regimens. rAd-p53 is a replication-defective adenoviral vector delivering the wild-type human TP53 gene to tumor cells, aiming to restore or augment p53 tumor suppressor function. The mechanism involves induction of cell cycle arrest, apoptosis, increased chemosensitivity, inhibition of angiogenesis, immune activation against tumor cells, and reduction of chemoresistance. When used in combination with chemotherapy, rAd-p53 has demonstrated improved tumor response rates and survival outcomes in several cancers (notably cervical cancer and hepatocellular carcinoma) compared to chemotherapy alone. This therapy is approved for certain cancers in China and is under investigation in other indications[1][2][3][4][5].
