Overview

rAd.p21 is a recombinant adenovirus-based gene therapy vector encoding the human p21 gene (also known as CDKN1A), a cyclin-dependent kinase inhibitor involved in cell cycle regulation, senescence, and apoptosis. By delivering the p21 gene, rAd.p21 aims to induce overexpression of the p21 protein, resulting in G1 cell cycle arrest, inhibition of fibroblast proliferation, and attenuation of pathological tissue proliferation. The therapy has been investigated as an adjunctive anti-proliferative treatment to prevent scarring and failure following glaucoma filtering surgeries, as well as in other contexts where limiting abnormal cell growth is desired. Subconjunctival injection localizes expression primarily to ocular tissues. Mechanistically, p21 inhibits cyclin-CDK2 and -CDK4 complexes, regulates the DNA polymerase co-factor PCNA, and is a downstream effector of p53-mediated cell cycle arrest[1][3][5][7].
