rAd26 (recombinant adenovirus type 26) is a replication-deficient viral vector used as a primary component of the COVID-19 vaccine Gam-COVID-Vac (Sputnik V) and as the sole active ingredient in the single-dose vaccine Sputnik Light. Developed by the Gamaleya National Research Centre of Epidemiology and Microbiology, the vector is engineered to carry the gene encoding the full-length spike (S) glycoprotein of the SARS-CoV-2 virus. In the heterologous prime-boost Sputnik V regimen, rAd26 is administered as the first dose to initiate an immune response, followed by a second dose using a different vector (rAd5) to bypass potential anti-vector immunity. The vaccine induces both neutralizing antibodies and T-cell mediated immunity against the SARS-CoV-2 virus.
