RAD301 (also known as NM-02) is a radiopharmaceutical candidate consisting of a nanobody (single-domain antibody) that specifically targets Protein Tyrosine Kinase 7 (PTK7). PTK7 is a pseudo-kinase receptor that is highly overexpressed in a variety of solid tumors, including triple-negative breast cancer (TNBC), non-small cell lung cancer (NSCLC), and colorectal cancer, while maintaining low expression in healthy tissues. Developed through a licensing agreement between NanoMab and Radiopharm Theranostics, RAD301 is designed as a theranostic agent, capable of being labeled with radioisotopes for both diagnostic imaging (e.g., Technetium-99m) and targeted radionuclide therapy. The nanobody modality offers advantages over traditional antibodies, such as smaller size for better tumor penetration and rapid systemic clearance, which enhances the imaging contrast and potentially reduces off-target radiation exposure.
