rAd35 Env A is a recombinant, replication-deficient adenovirus serotype 35 vector-based vaccine encoding the HIV envelope glycoprotein (Env) from clade A. It was developed as a candidate preventive HIV vaccine to stimulate both humoral and cellular immune responses against HIV. The rationale for using the Ad35 vector is its low seroprevalence in human populations, which may circumvent pre-existing immunity that can limit the effectiveness of more common vectors like Ad5. In clinical studies, rAd35 Env A was well tolerated and immunogenic, especially when used in heterologous prime/boost regimens with other adenoviral vectors such as rAd5[1][3]. The primary mechanism of action is immunostimulation—eliciting both antibody and T cell responses specific to the encoded HIV envelope antigen.
