Overview

rAd5 is a recombinant, replication-defective human adenovirus serotype 5 (Ad5) viral vector. It is engineered to be unable to replicate in normal host cells and is widely used as a platform for vaccine development and gene therapy. The rAd5 vector can deliver genetic material encoding antigens from various pathogens or therapeutic genes into host cells, leading to robust immune responses—particularly strong CD8+ T cell responses—making it valuable for vaccines against infectious diseases (such as HIV, influenza, rabies, Ebola, SARS-CoV-2) and as an oncolytic agent in cancer therapy. Its mechanism of action involves transducing target cells with the encoded antigen or gene of interest; the expressed protein then stimulates both humoral and cellular immunity[3][4][5]. The platform has been developed by multiple research groups and companies globally.
