rAd5-EBV-LMP2 is a therapeutic vaccine candidate designed to treat Epstein-Barr virus (EBV)-associated cancers, particularly nasopharyngeal carcinoma (NPC). It utilizes a recombinant serotype 5 adenoviral vector (rAd5) to deliver the gene encoding EBV latent membrane protein 2 (LMP2), an antigen implicated in NPC progression. The vaccine aims to stimulate antigen-specific cellular immunity by eliciting cytotoxic T lymphocyte responses against LMP2-expressing tumor cells. In phase I clinical trials, the vaccine was administered intramuscularly and demonstrated good tolerability with mostly mild adverse events such as fatigue, myalgia, cough, and injection site tenderness. Immunologically, it increased the proportion of CD3+CD4+ T cells in peripheral blood at higher doses. The high dose regimen is recommended for further study in phase II trials[1][6][9].
