A recombinant adenovirus serotype 5 (rAd5) viral vector vaccine designed to express the HIV-1 Clade B envelope (Env) protein. Developed by the Vaccine Research Center (VRC) of the National Institute of Allergy and Infectious Diseases (NIAID), this vaccine is part of a strategy to develop a preventive HIV vaccine using prime-boost regimens. The rAd5 vector is engineered to be replication-deficient and serves as a delivery vehicle for the HIV-1 gene insert. In clinical trials such as HVTN 083 (NCT01095224), it has been studied in combination with other vectors, such as rAd35, to evaluate the safety and immunogenicity of heterologous-insert and heterologous-vector prime-boost schedules. The goal of these regimens is to elicit broad T-cell and antibody responses against multiple HIV-1 subtypes.
